NVP-BEZ235 is a dual inhibitor of class I PI3Ks (IC50s = 4, 75, 7, and 5 nM, for p110α, - β, -δ, and -γ, respectively) and mTOR (IC50 = 20 nM).{18570} It is selective for these enzymes over a panel of kinases (IC50s = >10 µM for all). NVP-BEZ235 (0.1 or 1 µM) inhibits proliferation of, and induces apoptosis in, MCF-7, MDA-MB-468, SK-BR-3, and MDA-MB-361 breast cancer cells.{18572} It reduces tumor growth in a U87MG mouse xenograft model when administered at a doses of 25, 35, or 45 mg/kg.{18570}
