Ganaxolone is a positive allosteric modulator of GABAA receptors and a derivative of the neurosteroid allopregnanolone.{64380} It enhances GABA-induced currents in Xenopus oocytes expressing human recombinant ?1?1?2L, ?2?1?2L, ?3?1?2L subunit-containing GABAA receptors (EC50s = 213, 94, and 122 nM, respectively). Ganaxolone inhibits seizures induced by pentylenetetrazole (PTZ; Item No. 18682), bicuculline (Item No. 11727), aminophylline (Item No. 22235), or tert-butylbicyclophosphorothionate (TBPS) in mice (ED50s = 4.3, 4.6, 11.5, and 11.7 mg/kg, respectively).
