Product Overview:
This nuclear receptor assay system uses a specific rodent cell line that is further modified to express the mouse/rat peroxisome proliferator-activated receptor gamma (NR1C3, PPARG). Because the receptor’s ligand binding domain sequence is conserved between mouse and rat species, the receptor is denoted herein as mrPPARγ. These mrPPARγ reporter cells incorporate a responsive luciferase reporter gene, therefore, quantifying expressed luciferase activity provides a sensitive surrogate measure of mrPPARγ activity in the treated cells. The principal application of this reporter assay system is in the screening of test samples to quantify any functional activity, either agonist or antagonist, that they may exert against mrPPARγ. mrPPARγ reporter cells are prepared using Indigo’s specific CryoMite™ process. This cryo-preservation method yields high cell viability post-thaw, and provides the convenience of immediately dispensing healthy, division-competent reporter cells into assay plates. There is no need for intermediate spin-and-wash steps, viability determinations, or cell titer adjustments. Indigo Bioscience’s nuclear receptor reporter assays are all-inclusive cell-based assay systems. In addition to mrPPARγ reporter cells, this kit provides two optimized media for use in thawing the frozen cells and in diluting the user’s test samples, a reference agonist, luciferase detection reagent, and a cell culture-ready assay plate. Test compounds can be screened for agonist or antagonist activities against human nuclear receptors expressed within the cytoplasm and nuclear environments of healthy, dividing mammalian cells.

Our reporter systems utilize fire fly luciferase reporter gene technology to provide optimal assay sensitivity and dynamic range when quantifying nuclear receptor activity. All of INDIGO Biosciences’ nuclear receptor reporter assay systems incorporate a detection reagent specially formulated to provide stable light emission over at least I hour, allowing users to dispense detection reagent into all assay wells prior to commencing activity measurements. This eliminates the need for a luminometer equipped with injectors, allows plates to be processed in batch, and dramatically reduces the start-to-finish read-time of assay plates.

INDIGO reporter assays are optimally configured for processing in 96- and 384-well format assay plates. Single plate kits (3x 32-, 1x96-, and 1x 384-well) are offered as all-inclusive assay systems. Each kit includes nuclear receptor reporter cells, an assay plate, all required reagents (optimized culture media, positive-control agonist, and detection reagent), as well as a detailed Technical Manual.
